    1. Add information about the experiment conditions by clicking on the blue Conditions button in the figure dimensions section . A "Conditions" box will appear.
    2. Click on the Click to Edit button in the conditions box to get to the condition annotations.
    3. In the "Create conditions" text area, type "Unstim, IL-4, IFNg, IL-4 + IFNg, G-CSF, IFN Type I, IL-6" and click Add.

      Under "Select a condition", each condition tag will now have its own tab. (You may have to wait a moment until the page is finished loading.)

    4. Cytobank tries to assign tags based on the FCS filename and the FCS sample name field. Sometimes the terminology will be slightly different on the file; it's easy to drag-and-drop to move the files into the correct tag. Look under "Untagged"; you will see "2-4) U937 IFN-g". Cytobank couldn't sort that because there is a hyphen between "IFN" and "g", unlike the tag you typed in.
    5. Click on the tab for IFNg. Now you should see the "Untagged" column and the "IFNg Tagged Files" column. This can make it easier to move files than using the "All Conditions" view. Drag and drop the "2-4) U937 IFN-g" file into the proper "IFNg Tagged Files" column.
    6. Try changing the tag name by double-clicking on IFNg just under the "All Conditions" tab; a text field will appear. Change it to "IFN-g" and press Enter.
    7. Click on All Conditions. Under "IL-4", you will notice "2-5) U937 IL-4 + IFN-g". Again, the hyphen in "IFN-g" has confused things. Drag-and-drop this file into the "IL-4 + IFNg" column.
    8. Check that all the other files look like they are in the correct columns. They should be.
    9. When you feel that everything is sorted correctly, click on Return to Illustration.
    10. You will see the Working Illustration page. The condition tags you just created will be in the "Conditions" box.
